Ratio, PharmaLogic expand Ac-225 manufacturing collaboration

Ratio Therapeutics, a radiopharmaceutical developer based in Boston, Massachusetts, has expanded its manufacturing collaboration with PharmaLogic, a contract development and manufacturing organization (CDMO) specializing in radiopharmaceuticals, to support continued clinical development and commercial readiness planning for its lead candidate, RTX-2358, an Actinium-225-based therapy targeting fibroblast activation protein-alpha (FAP).

Under the expanded agreement, PharmaLogic will increase manufacturing capacity for clinical supply of RTX-2358 by adding production at its Idaho Falls, Idaho therapeutics facility. The collaboration will also include enhancements to production processes to enable larger-scale, multi-dose batch manufacturing, the company said. The partnership is designed to support ongoing and future clinical studies, including potentially registrational trials evaluating RTX-2358 in FAP-expressing tumors, according to the announcement.

Jack Hoppin, CEO of Ratio Therapeutics, said in a statement that reliable, redundant, and scalable manufacturing infrastructure is critical to the successful development of Ac-225-based therapies, and that the expanded collaboration is intended to support clinical trial progression and commercial supply readiness.

Etienne Montagut, president and CEO of PharmaLogic, added that the addition of the Idaho Falls facility and enhanced batch production capabilities will provide the capacity, redundancy, and scalability the RTX-2358 program requires as it advances toward commercialization.

The announcement comes amid growing activity across the Ac-225 supply chain. Last week, NorthStar Medical Radioisotopes and QSA Global announced a multi-year agreement to establish a closed-loop system for processing and recycling Radium-226 — the material from which Ac-225 is produced — to improve supply reliability for targeted alpha therapy manufacturers. In addition, Niowave recently broke ground on a $75 million Ac-225 production facility in Lansing, Michigan, expected to begin operations in 2028.

In April, PharmaLogic opened a new PET radiopharmaceutical manufacturing facility in Atlanta, Georgia. The site is designed to expand domestic production capacity for diagnostic and therapeutic radiopharmaceuticals, while supporting research and development of next-generation products.
